The 25-year-old Pole, ranked world No. 14 by the Association of Tennis Professionals (ATP), dispatched Serbian player Dušan Lajović in straight sets, 7-5, 6-3, in their third-round encounter on Thursday.
A day earlier, Hurkacz defeated home favourite Alejandro Davidovich Fokina, 6-4, 4-6, 7-6, in the second round.
Up next for the Polish player at the clay-court tournament in the Spanish capital is Serbia's world No. 1 Novak Djokovic.
Meanwhile, Poland's Iga Świątek, the world No. 1 in women's tennis, said last month she would miss the Madrid Open due to a shoulder injury.
The 2022 Madrid Open runs until Sunday.
